目的研究从中药槲寄生中制备的槲寄生凝集素蛋白诱导肿瘤细胞凋亡机制。方法用经典的染色技术、流式细胞术、MTT法分析经分子生物学方法分离纯化的槲寄生凝集素蛋白诱导肿瘤细胞凋亡形态特征,AP峰值和抑制肿瘤细胞增殖的抑制率以研究其诱导肿瘤细胞凋亡机制。结果槲寄生凝集素蛋白具有诱导肿瘤细胞凋亡和显著抑制和阻断肿瘤细胞增殖且以Caspase依赖的方式发挥其生物学作用的。结论槲寄生凝集素蛋白具有直接抗肿瘤作用,在肿瘤免疫学的基础和临床应用研究中具有前景。
Objective To study the mechanism of tumor cell apoptosis induced by the mistletoe lectin protein prepared from traditional Chinese medicine mistletoe. Methods To analyze the morphological features of apoptotic cells, the peak value of AP and the inhibitory rate of inhibiting the proliferation of tumor cells by classical staining technique, flow cytometry and MTT assay to study the induction of the mistletoe lectin protein by molecular biology method Tumor cell apoptosis mechanism. Results Mistletoe lectin protein can induce tumor cell apoptosis and significantly inhibit and block tumor cell proliferation and play its biological role in a Caspase-dependent manner. Conclusion Mistletoe agglutinin protein has a direct anti-tumor effect, and has a bright prospect in the basic and clinical application of tumor immunology.
