目的建立指纹图谱与主成分分析(PCA)相结合的提取工艺优化方法,并将其应用于复方虎杖方乙醇提取工艺优化。方法以复方虎杖方为模型药物,采用HPLC法建立指纹图谱,对共有峰峰面积进行PCA,以总因子得分为评价指标,采用U9(93×31)均匀设计法考察提取次数(X1)、乙醇用量(X2)、乙醇体积分数(X3)和提取时间(X4)对提取效果的影响,建立回归模型,确定最佳工艺参数。结果建立的数学模型能够较好地描述效应指标与各因素之间的关系(r=0.997),应用建立的方法优化得到复方虎杖方最适宜醇提工艺为6倍量90%乙醇提取3次,每次1 h。结论所建立的方法简便有效,适合复方虎杖方提取,可以为复方中药提取工艺优化提供参考。
OBJECTIVE To establish a method for the optimization of extraction process combining fingerprint and principal component analysis (PCA) and apply it to the optimization of ethanol extraction process of compound Polygoni cuspidatum. Methods The compound prescription of Huzhang Fang was used as model drug. HPLC method was used to establish the fingerprint and the total peak area was determined by PCA. The total factor score was used as the evaluation index. U9 (93 × 31) uniform design method was used to investigate the extraction times (X1) The effects of dosage (X2), ethanol volume fraction (X3) and extraction time (X4) on the extraction efficiency were studied. A regression model was established to determine the optimum process parameters. Results The mathematical model established can well describe the relationship between the effect index and each factor (r = 0.997). The optimal alcohol extraction process was obtained by 6 times 90% 1 h each. Conclusion The established method is simple and effective. It is suitable for the compound prescription of Polygoni cuspidatum and can provide a reference for the extraction process optimization of Chinese herbal medicine.
