目的 探讨γ射线对大鼠血管平滑肌细胞 (VSMCs)抑制作用的机理。方法 应用克隆形成实验绘制VSMCs剂量存活曲线 ,采用3H掺入法观察γ射线对VSMCs增殖的影响。应用流式细胞仪和“DNALadder”技术检测γ射线对VSMCs凋亡的影响。结果 VSMCs的D0 值为 1 95Gy,Dq值为 1 76Gy ,D37值为 3 71Gy ,N值为 2 47。射线对VSMCs增殖的抑制作用呈剂量依赖性 ,随剂量增加而增大。 2、10、2 0Gy照射后 48h内VSMCs凋亡数量均未增加。结论 γ射线可抑制VSMCs增殖 ,其机理主要是γ射线对VSMCs中有克隆形成能力的细胞进行杀伤以及抑制其进行有丝分裂。
Objective To investigate the mechanism of the inhibitory effect of γ-rays on rat vascular smooth muscle cells (VSMCs). Methods The dose-dependent survival curves of VSMCs were drawn by clonogenic assay. The effects of γ-rays on the proliferation of VSMCs were observed by 3H incorporation. The effects of γ-rays on VSMCs apoptosis were detected by flow cytometry and “DNALadder” technique. Results VSMCs had a D0 value of 1 95Gy, a Dq value of 1 76Gy, a D37 value of 3 71Gy and an N value of 2 47. The inhibitory effect of radiation on the proliferation of VSMCs in a dose-dependent manner, with the dose increased. The number of apoptotic VSMCs did not increase within 48h after 2,10,2 OGy irradiation. Conclusion γ-rays can inhibit the proliferation of VSMCs. The main mechanism is γ-ray killing of clonogenic cells in VSMCs and their mitosis.
