目的观察脑出血血肿周围组织T细胞免疫球蛋白黏蛋白分子(TIM)-3的表达,并探讨其在脑出血发病中的意义。方法行血肿清除术的脑出血患者48例按发病距手术取材时间分为≤6 h组、6~24 h组、24~72 h组和>72 h组,每组12例。另选取远离血肿的脑组织12例作为对照组。免疫组织化学染色法检测TIM-3蛋白的表达,RT-PCR法检测TIM-3 m RNA的表达。结果与对照组相比,≤6 h组血肿周围组织中TIM-3的表达无统计学差异(P>0.05)。与对照组相比,其他各实验组血肿周围组织中TIM-3的表达均具有统计学差异(P<0.05)。24~72 h组血肿周围组织中TIM-3的表达显著高于其他各实验组(P<0.05)。RT-PCR法检测血肿周围组织TIM-3的表达表现出相同的趋势。结论脑出血血肿周围脑组织TIM-3的表达在发病6 h后增高,24~72 h达最高,72 h后降低。TIM-3参与脑出血后的病理过程,可能与炎症反应有关。
Objective To observe the expression of T cell immunoglobulin mucin molecule (TIM-3) in perihematomal hematoma and to explore its significance in the pathogenesis of cerebral hemorrhage. Methods Forty-eight patients with intracerebral hemorrhage who underwent hematoma removal were divided into 6 hours group, 6-24 hours group, 24-72 hours group and> 72 hours group according to the onset time of operation. Twelve cases in each group. Another 12 cases of brain tissue away from hematoma as a control group. The expression of TIM-3 protein was detected by immunohistochemical staining, and the expression of TIM-3 mRNA was detected by RT-PCR. Results Compared with the control group, there was no significant difference in the expression of TIM-3 in the perihematomal tissue between ≤ 6 h groups (P> 0.05). Compared with the control group, the expression of TIM-3 in the perihematoma tissue of other experimental groups was significantly different (P <0.05). The expression of TIM-3 in the perihematomal tissue around the 24-72 h group was significantly higher than that of other experimental groups (P <0.05). RT-PCR method to detect TIM-3 expression in the perihematoma showed the same trend. Conclusions The expression of TIM-3 in brain tissue around hematoma of ICH increased at 6 h after onset, reaching the highest at 24 ~ 72 h and then decreased at 72 h. TIM-3 involved in the pathological process of cerebral hemorrhage may be related to the inflammatory response.
