为了解胰高血糖素和双丁酰环腺苷酸在肝脏载脂蛋白代谢中的作用 ,以培养的人肝癌细胞系HepG2细胞为研究对象 ,采用“冻干浓缩培养液载脂蛋白测定法” ,观察了胰高血糖素和双丁酰环腺苷酸对HepG2细胞分泌载脂蛋白AⅠ、AⅡ、CⅢ、B10 0及E的影响。结果表明 ,胰高血糖素浓度为 4× 10 -7mol/L时 ,载脂蛋白AⅠ、AⅡ、B10 0及E的分泌分别减少 47.7%、5 4.1%、32 .7%和 33.2 % (P <0 .0 1) ,载脂蛋白CⅢ的分泌增加 89.1% (P <0 .0 1) ;双丁酰环腺苷酸的浓度为 1× 10 -3mol/L时 ,载脂蛋白AⅠ、B10 0和E的分泌分别减少 12 .3 %、2 4.9%和 16 .1%(P <0 .0 1) ,载脂蛋白AⅡ和CⅢ的分泌分别增加 6 6 .1%和 142 .6 % (P <0 .0 1)。胰高血糖素和双丁酰环腺苷酸对HepG2细胞载脂蛋白分泌的影响基本一致。
In order to understand the role of glucagon and dibutyryl cyclic adenosine monophosphate in the hepatic apolipoprotein metabolism, HepG2 cells cultured in human hepatocellular carcinoma cell line HepG2 as the research object, using “freeze-dried concentrated culture apolipoprotein assay” to observe The effects of glucagon and dibutyryl cyclic adenosine monophosphate on the secretion of apolipoprotein AⅠ, AⅡ, CⅢ, B10 0 and E in HepG2 cells were studied. The results showed that the secretion of apolipoproteins AⅠ, AⅡ, B10 0 and E decreased by 47.7%, 5 4.1%, 32.7% and 33.2%, respectively, at the concentration of 4 × 10 -7 mol / L (P < (P <0.01).
