为筛选出适宜何首乌组织培养的培养基,以带腋芽茎段为外殖体,采用完全随机区组试验设计,进行不同浓度激素(6-BA,NAA,IBA,6-BA+NAA,6-BA+IBA,多效唑)的培养基对何首乌产生丛生芽及不定根的诱导作用研究。结果表明:一定浓度范围内6-BA对诱导何首乌产生丛生芽具有促进作用,NAA和IBA对丛生芽的诱导具有抑制作用,适宜何首乌丛生芽增殖的培养基为MS+6-BA 1.0mg/L,增殖系数达11.43;多效唑对不定根的诱导具有促进作用,最适生根的培养基为1/2MS+多效唑0.5mg/L,生根率为100%,平均生根条数为10.42条/株。
In order to select suitable culture medium for Polygonum multiflorum Thunb. Culture, the stem segments with axillary buds were used as explants. Randomized block design with different concentrations of hormones (6-BA, NAA, IBA, 6-BA + NAA, BA + IBA, paclobutrazol) medium on Polygonum multiflorum buds and adventitious roots induced role. The results showed that 6-BA could promote the production of polygonum multiflorum in a certain concentration range, while NAA and IBA could inhibit the induction of clustered buds. MS medium supplemented with 6-BA 1.0 mg / L , And the multiplication coefficient reached 11.43. The effect of paclobutrazol on adventitious root induction was promoted. The optimum medium for rooting was 1 / 2MS + paclobutrazol 0.5mg / L, the rooting rate was 100% and the average number of rooting was 10.42 / plant.
