目的检测树突状细胞(DC)及其各亚群[髓样树突状细胞(mDC)和浆细胞样树突状细胞(pDC)]在结核性胸膜炎(TP)患者外周血及胸腔积液中的变化,并且分析其与白细胞介素-12(IL-12)水平的相关性,探讨其临床意义。方法以12例健康志愿者作为对照组,采用流式细胞术对16例TP患者(TP组)外周血和胸腔积液中总体树突状细胞(tDC)及其各亚群的比例进行检测;同时,用ELISA法对TP患者外周血浆和胸腔积液中IL-12的含量进行检测。结果 TP组患者外周血中tDC和mDC比例显著高于对照组,治疗后tDC比例下降明显;TP组患者胸腔积液中tDC和mDC比例高于外周血,但pDC低于外周血;TP组患者血浆中IL-12的含量显著高于对照组,而胸腔积液中IL-12的含量则明显高于其血浆含量;TP患者血浆中IL-12的水平与外周血tDC、mDC呈正相关,并且胸腔积液中IL-12的水平也与胸腔积液tDC、mDC呈正相关。结论DC,尤其是mDC在TP中具有重要的抗结核免疫作用。
Objective To detect the expression of dendritic cells (DCs) and their subpopulations (myeloid dendritic cells (mDC) and plasmacytoid dendritic cells (pDCs)] in peripheral blood and pleural effusion of patients with tuberculous pleurisy (TP) , And to analyze its correlation with interleukin-12 (IL-12) levels to explore its clinical significance. Methods Twenty - two healthy volunteers served as control group, and the proportion of total dendritic cells (tDC) and its subsets in peripheral blood and pleural effusion of 16 TP patients (TP group) was detected by flow cytometry. At the same time, the content of IL-12 in peripheral blood plasma and pleural effusion of TP patients was detected by ELISA. Results The proportion of tDC and mDC in peripheral blood of TP group was significantly higher than that of control group, and the percentage of tDC decreased significantly after treatment. The proportion of tDC and mDC in pleural effusion of TP group was higher than that of peripheral blood, but pDC was lower than that of peripheral blood. The content of IL-12 in plasma was significantly higher than that in control group, while the content of IL-12 in pleural effusion was significantly higher than that in plasma. The level of IL-12 in plasma of TP patients was positively correlated with tDC and mDCs in peripheral blood The level of IL-12 in pleural effusion was also positively correlated with tDC and mDC in pleural effusion. Conclusions DC, especially mDC, has an important anti-TB immune effect in TP.
