目的探讨人12周龄胚胎脑脉络丛上皮细胞是否具有神经干细胞的生物学特性。方法制备室管膜/室管膜下区和纹状体脑切片和脉络丛组织铺片,采用免疫荧光染色,在激光扫描共焦显微镜下观察结果,采集图像数据。结果人12周龄胚胎脑室脉络丛上皮细胞表达神经干细胞分子标志CD133、Nestin和Sox2。神经干细胞的分子标志在脉络丛上皮细胞的分布具有一定极性,未见共定位分布。室管膜上皮细胞面向脑室腔面的细胞顶端微绒毛也表达CD133,而室管膜下区和纹状体区可见极少数CD133阳性标记细胞。结论 12周龄脑室脉络丛上皮细胞已定向分化,不仅提供维持神经干细胞的微环境,还具有神经干细胞的生物学特性。
Objective To investigate whether human 12-week-old embryonic choroid plexus epithelial cells have the biological characteristics of neural stem cells. Methods Ependymal / subventricular zone and striatum brain slice and choroid plexus were prepared. The immunofluorescence staining was used to observe the results under laser scanning confocal microscope and the image data was collected. Results Human 12-week-old embryonic ventricular choroid plexus epithelial cells expressed neural stem cell markers CD133, Nestin and Sox2. The distribution of neural stem cells molecular markers in choroid plexus epithelial cells has a certain polarity, no co-localization distribution. The ependymal cells of the ependymal epithelial cells also express CD133 on the apical microvilli of the cells, while very few CD133 positive labeled cells can be seen in the subependymal and striatal areas. Conclusion 12-week-old ventricular choroid plexus epithelial cells have been oriented differentiation, not only to maintain the microenvironment of neural stem cells, but also has the biological characteristics of neural stem cells.
