目的探讨电磁辐射对体外培养的淋巴细胞(Raji细胞)超微结构及其培养上清液中TNF-α水平的影响。方法体外无血清培养Raji细胞,分别用电磁脉冲(EMP)、S波段高功率微波(S-HPM)、X波段高功率微波(X-HPM)为辐射源辐射对数期生长的Raji细胞,采用电镜技术观察辐射6 h时Raji细胞超微结构的改变,各组随机观察100个细胞,通过计数其中受损细胞数对损伤的细胞进行半定量;采用放射免疫分析法检测辐射6 h时Raji细胞培养上清液中TNF-α的水平;所得数据分别采用SPSS 13.0统计学软件的χ2、t检验进行统计学分析,以P<0.05具有统计学意义。结果与对照组相比,各实验组Raji细胞于辐照后6 h超微结构均发生不同程度破坏,损伤的程度以S-HPM最轻,仅表现为线粒体、内质网和核膜结构轻度受损,以X-HPM最重,除细胞器结构严重破坏外,见凋亡小体形成,EMP的损伤程度介于S-HPM和X-HPM之间;半定量结果:与对照组比较,损伤的细胞数S-HPM组最少(P<0.01),X-HPM组最多(P<0.01),EMP组居中间(P<0.01),各实验组间差异显著(P<0.01或0.05)。放射免疫分析法未检测到Raji培养上清液中TNF-α水平的改变(P值均>0.05)。结论 EMP、S-HPM和X-HPM三种电磁辐射6 h均可损伤Raji细胞,导致其超微结构的破坏乃至凋亡坏死,损伤程度与电磁波的种类有关,但不影响Raji细胞培养上清液中TNF-α的水平。
Objective To investigate the effects of electromagnetic radiation on the ultrastructure of lymphocytes (Raji cells) cultured in vitro and the level of TNF-α in culture supernatant. Methods Raji cells were cultured in serum-free medium. Raji cells were irradiated with electromagnetic pulse (EMP), S-HPM and X-HPM respectively. The ultrastructural changes of Raji cells were observed under electron microscope at 6 hours. 100 cells were randomly observed in each group, and the number of damaged cells was counted by semi-quantitative method. Radioimmunoassay was used to detect Raji cells The level of TNF-α in the supernatant was cultured. The data were statistically analyzed by χ2, t test using SPSS 13.0 statistical software, respectively, with statistical significance at P <0.05. Results Compared with the control group, the ultrastructure of Raji cells in each experimental group were damaged to varying degrees 6 h after irradiation, and the degree of injury was the lightest in S-HPM, showing only mitochondria, endoplasmic reticulum and nuclear membrane structure The degree of damage of EMP was between S-HPM and X-HPM. Semiquantitative results: Compared with the control group, The number of injured cells in the S-HPM group was the lowest (P <0.01), that in the X-HPM group was the highest (P <0.01), and that in the EMP group was intermediate (P <0.01). Radioimmunoassay did not detect changes in TNF-α levels in Raji culture supernatants (P> 0.05). Conclusion EMP, S-HPM and X-HPM all can damage Raji cells at 6 h, resulting in the destruction of ultrastructure and apoptosis necrosis. The degree of damage is related to the type of electromagnetic wave but does not affect Raji cell culture supernatant Fluid TNF-α levels.
