目的 :探讨芦荟大黄素抑制平滑肌细胞增殖的作用机制。方法 :用 3FFogarty球囊拉栓导管对纯种日本大耳白兔腹主动脉进行球囊内皮剥脱损伤 ,4 8h后取出腹主动脉中膜进行原代平滑肌细胞培养 ,细胞同步于G0 /G1期后 ,实验组加芦荟大黄素 2 0 μg/ml,对照组加等体积的细胞培养液 ,3h后分别用North ern杂交法、Western杂交法和免疫细胞化学技术检测两组动物平滑肌细胞c myc基因mRNA和蛋白表达产物的变化。结果 :与对照组比较 ,实验组平滑肌细胞c mycmRNA和蛋白水平的表达变化差异均无显著性。结论 :芦荟大黄素对平滑肌细胞的增殖抑制作用可能不是通过影响c myc基因的表达实现的 ,而是通过其他途径。
Objective : To explore the mechanism of action of aloe-emodin in inhibiting the proliferation of smooth muscle cells. METHODS: Abdominal aorta of Japanese rabbits were subjected to balloon endothelium injury with a 3FFogarty balloon catheter. The abdominal aorta medial membrane was removed after 48 hours for primary smooth muscle cell culture. The cells were synchronized to G0/G1 phase. After the experiment, aloe-emodin 20 μg/ml was added to the experimental group. The control group was added with an equal volume of cell culture medium. After 3 hours, the myc gene of smooth muscle cells was detected by Northern hybridization, Western blot and immunocytochemistry. Changes in mRNA and protein expression products. Results: Compared with the control group, the expression changes of c myc mRNA and protein levels in the smooth muscle cells of the experimental group were not significantly different. Conclusion : The inhibitory effect of aloe-emodin on the proliferation of smooth muscle cells may not be achieved by affecting the expression of c myc gene, but by other means.
