目的 :在临床研究和动物实验的基础上 ,进一步探讨健骨二仙丸对成骨细胞增殖和细胞周期的影响。方法 :分离、培养人的原代成骨细胞 ,采用3 H 胸腺嘧啶掺入法和四氮唑蓝法检测细胞增殖 ,流式细胞仪检测细胞周期。结果 :健骨二仙丸中、高剂量组细胞增殖率、S期细胞比率和细胞增殖指数等均显著高于空白对照组 (P <0 0 1,P <0 0 5 ) ,与雌激素组比较差异无显著性。细胞凋亡率和G0 G1期细胞比率显著低于空白对照组 (P <0 0 1,P <0 0 5 )。结论 :健骨二仙丸能有效促进成骨细胞的增殖、分化 ,防止成骨细胞的凋亡
Objective : To further investigate the effects of Jiangu Erxian Pill on the proliferation and cell cycle of osteoblasts on the basis of clinical studies and animal experiments. METHODS: Human primary osteoblasts were isolated and cultured. Cell proliferation was detected by 3 H thymidine incorporation and tetrazolium blue. Cell cycle was measured by flow cytometry. RESULTS: The proliferation rate, S-phase cell ratio and cell proliferation index in the medium and high dose groups of Jiangu Erxian Pills were significantly higher than those in the blank control group (P < 0.01, P <0.05), and the estrogen group. The difference was not significant. The cell apoptosis rate and G0 G1 phase cell ratio were significantly lower than those in the blank control group (P <0 01, P <0 05). Conclusion: Jiangu Erxian Pill can effectively promote the proliferation and differentiation of osteoblasts and prevent the apoptosis of osteoblasts.
