目的:观察白英总生物碱的抗炎活性。方法:培养人脐静脉内皮细胞(HUVEC),用200μmol·L~(-1)双氧水(H_2O_2)损伤细胞;培养小鼠单核/巨噬细胞(RAW 264.7),用脂多糖(LPS)诱导;抽签法随机分成正常组、模型组、阳性对照组、白英总生物碱低、中、高剂量组。给药后,继续培养,应用CCK-8法观察细胞存活率;饲养SD大鼠,抽签法随机分为正常组、模型组、阳性对照组、白英总生物碱低、中、高剂量组,观察白英总生物碱对角叉菜胶诱导大鼠急性踝关节肿胀度的影响,测定前列腺素E2(PGE_2)、环氧化物水解酶-2(COX-2)含量。结果:白英总生物碱中剂量对H_2O_2损伤的HUVEC细胞的影响以及高剂量组对LPS诱导的巨噬细胞RAW 264.7的影响,均和阳性对照组相当,差异无统计学意义(P>0.05);中、高浓度白英总生物碱能够明显降低大鼠急性踝关节肿胀度,降低大鼠足趾渗出液中PGE2、血清中COX-2含量,与模型组差异有统计学意义(P<0.01)。结论:白英总生物碱具有明显的抗炎活性,有必要进一步深入研究其药效及作用机制。
Objective: To observe the anti-inflammatory activity of total alkaloids of Crab White Methods: Human umbilical vein endothelial cells (HUVECs) were cultured and injured with 200μmol·L -1 H 2 O 2. RAW 264.7 cells were cultured and induced by lipopolysaccharide (LPS) Lottery randomly divided into normal group, model group, positive control group, white British total alkaloids low, medium and high dose group. After administration, the cell survival rate was observed by CCK-8 method. SD rats were bred and randomly divided into normal group, model group, positive control group, low, middle and high doses of total alkaloids To observe the effect of total alkaloids of Cortex Bletillae on carrageenan-induced acute ankle swelling in rats, and to determine the content of prostaglandin E2 (PGE_2) and cyclooxygenase-2 (COX-2). Results: The effect of middle dose of total alkaloids on HUVEC cells injured by H 2 O 2 and the effect of high dose group on LPS-induced RAW 264.7 macrophages were similar to that of the positive control group, but the difference was not statistically significant (P> 0.05) ; Middle and high concentrations of total white alkaloids can significantly reduce acute ankle swelling in rats, reduce rat foot exudate PGE2, serum COX-2 levels, and the model group difference was statistically significant (P < 0.01). Conclusion: The total alkaloids have a significant anti-inflammatory activity, it is necessary to further study its efficacy and mechanism.
