目的建立显齿蛇葡萄茎中黄酮类成分的含量测定方法,分析广西不同地区显齿蛇葡萄茎中二氢杨梅素、芦丁及杨梅素的含量。方法采用色谱柱为Poroshell EC-C_(18)(4.6 mm×250 mm,4.0μm),以乙腈-0.1%磷酸为流动相,梯度洗脱,流速1 m L·min~(-1),柱温25℃,检测波长0~23 min,330 nm;23~50 min,375 nm。结果显齿蛇葡萄茎中二氢杨梅素、芦丁及杨梅素进样量分别在0.800 5~9.606μg(r=0.999 9)、0.201 6~2.419μg(r=0.999 8)和0.100~1.202μg(r=0.999 7)内呈良好的线性关系;平均加样回收率分别为98.1%,101%和99.9%,RSD<3%,方法的稳定性、重复性和精密度均良好(RSD<3%)。桂林灵川的显齿蛇葡萄茎中二氢杨梅素、芦丁和杨梅素的含量均较高,武鸣双江和金秀互助村的显齿蛇葡萄茎中二氢杨梅素和杨梅素含量均较低,而武鸣双江的芦丁含量较高,柳州泗顶镇的芦丁含量较低。结论不同地区显齿蛇葡萄茎中二氢杨梅素、芦丁和杨梅素的含量相差较大。该方法结果准确、重复性好,可使目标成分与试样中其他共存组分达到较好地分离。
OBJECTIVE To establish a method for determination of flavonoids in stems of Dendrocostus serrata, and to analyze the content of dihydromyricetin, rutin and myricetin in the stems of Dendrocostus serrata in different areas of Guangxi. Methods The column was Poroshell EC-C 18 (4.6 mm × 250 mm, 4.0 μm) with gradient elution with acetonitrile-0.1% phosphoric acid as mobile phase at a flow rate of 1 mL · min -1 Temperature 25 ℃, detection wavelength 0 ~ 23 min, 330 nm; 23 ~ 50 min, 375 nm. Results The injection volumes of dihydromyricetin, rutin and myricetin in the stems of H. victus were 0.800 5 ~ 9.606 μg (r = 0.999 9), 0.201 6 ~ 2.419 μg (r = 0.999 8) and 0.100 ~ 1.202 μg (r = 0.999 7). The average recoveries were 98.1%, 101% and 99.9%, respectively. The RSD was less than 3%. The stability, repeatability and precision of the method were all good %). The content of dihydromyricetin, rutin and myricetin in the stems of Gracilaria spp. In Lingchuan of Guilin were all higher than that in the stems of Vitex negundo Low, while the content of rutin in Wuming Shuangjiang is higher, and the content of rutin in Siding Town, Liuzhou is lower. Conclusions The content of dihydromyricetin, rutin and myricetin in the stems of Dendrocostus serrata in different regions is quite different. The method is accurate and reproducible, which can separate the target components and other coexisting components in the sample better.
