目的:研究新疆维吾尔族和汉族妇女宫颈鳞癌组织中β2-微球蛋白(β2-MG)的表达及其与人乳头状瘤病毒(HPV)16亚型感染的关系。方法:采用免疫组化方法和聚合酶链反应(PCR)技术检测75例宫颈炎及127例宫颈鳞癌组织中β2-MG的表达和HPV16感染情况。结果:汉族、维吾尔族宫颈鳞癌组织中β2-MG阳性表达率分别为78.95%、53.93%,明显低于宫颈炎组织中的阳性表达率(93.48%、100.00%,P<0.05);维吾尔族宫颈鳞癌组织中β2-MG阳性表达率(53.93%)明显低于汉族(78.95%)(P<0.05)。维吾尔族宫颈鳞癌组织中β2-MG的表达与HPV16感染呈正相关(r=0.162,P<0.05)。结论:宫颈鳞癌组织中β2-MG表达量下降,且在维吾尔族和汉族间表达有差异,提示HPV感染的宫颈鳞癌组织由于β2-MG表达量下降,影响了主要组织相容性复合体(MHC)-Ⅰ类分子的正确组装,不能有效地呈递抗原,可能是肿瘤细胞逃逸免疫监视的一种机制。
Objective: To investigate the expression of β2-microglobulin (β2-MG) and its relationship with human papillomavirus (HPV) 16 subtype in patients with Uigur and Han nationality in Xinjiang. Methods: The expressions of β2-MG and HPV16 in 75 cases of cervicitis and 127 cases of cervical squamous cell carcinoma were detected by immunohistochemistry and polymerase chain reaction (PCR). Results: The positive rates of β2-MG in Han and Uygur cervical squamous cell carcinomas were 78.95% and 53.93%, respectively, which were significantly lower than those in cervicitis (93.48%, 100.00%, P <0.05) The positive rate of β2-MG in cervical squamous cell carcinoma (53.93%) was significantly lower than that in Han (78.95%) (P <0.05). The expression of β2-MG in Uigur cervical squamous cell carcinoma was positively correlated with HPV16 infection (r = 0.162, P <0.05). CONCLUSIONS: The expression of β2-MG in cervical squamous cell carcinoma is decreased, and the expression is different between Uygur and Han ethnicity. It suggests that the expression of β2-MG in cervical squamous cell carcinoma of the cervix decreases, affecting the expression of major histocompatibility complex (MHC) -I molecules correctly assembled, can not effectively present the antigen, may be a mechanism for tumor cell escape immune surveillance.
