目的观察GnRH-A制剂——抑那通(注射用醋酸亮丙瑞林)诱导的特发性性早熟小鼠动物模型与阴虚火旺证的相关关系及知柏地黄丸的干预作用。方法采用抑那通诱导NIH雌性小鼠建立特发性性早熟模型,观察该模型的体温、体重、血清环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、cAMP/cGMP的变化;以甲地孕酮为阳性对照药,观察知柏地黄丸对上述指标变化的影响。结果抑那通诱导的模型动物表现出体质量及卵巢重量增加,子宫重量亦有增加的趋势;高、低剂量的知柏地黄丸及甲地孕酮均能对抗抑那通所致动物体温升高和卵巢重量的增加;低剂量的知柏地黄丸及甲地孕酮均能对抗抑那通所致动物血清cAMP含量升高。此外,除正常对照组外,各组血清cAMP含量与体温及卵巢重量之间均呈正的直线相关;阴虚火旺证模型组动物血清cAMP含量与子宫重量之间亦呈正的直线相关。结论抑那通诱导的特发性性早熟模型同时表现出阴虚火旺证的特征表现。知柏地黄丸在抑制抑那通诱导的性成熟提前的同时,亦能拮抗其诱导的阴虚火旺证的特征表现。表明抑那通诱导的特发性性早熟模型与阴虚火旺证之间具有一定的相关性。
Objective To observe the relationship between GnRH-A preparations, idiopathic precocious puberty mouse model induced by lenatone (injection with leuprolide acetate) and yin-wang-huoxue syndrome and the intervention effect of Zhibai Dihuang Wan. Methods The model of idiopathic precocious puberty was established in NIH female mice induced by Yonatong. The changes of body temperature, body weight, serum cAMP, cGMP and cAMP/cGMP were observed. Megestrol was a positive control drug and the effect of Zhibai Dihuang Wan on the above changes was observed. The results showed that the model animals induced by lenatone exhibited an increase in body weight, ovary weight, and uterine weight; high and low doses of Zhibai Dihuang Wan and megestrol acetate were able to counteract the temperature increase in animals caused by lenatone. The increase in the height and the weight of the ovary; the low doses of Zhibai Dihuang Wan and megestrol acetate were able to counteract the increase in serum cAMP levels in animals caused by itraton. In addition, in addition to the normal control group, serum cAMP levels were positively correlated with body temperature and ovary weight in each group. The serum cAMP level in yin-wang-huo-wang syndrome model group was also positively correlated with uterine weight. Conclusion The idiopathic precocious puberty model induced by lenatone shows the characteristics of yin deficiency and fire syndrome. Zhibai Dihuang Pills can inhibit the growth of sex-induced sexual maturation, but at the same time can antagonize the characteristic manifestations of Yin-deficiency and Huo-wang syndrome induced by it. It shows that there is a certain correlation between the idiopathic precocious puberty model induced by Yinatong and Yin deficiency syndrome.
