目的探讨静止压力对血管平滑肌细胞增殖与凋亡的影响。方法将胚胎鼠血管平滑肌细胞系A10细胞分别在正常大气压下、80、100、200mmHg等静止压力下培养;利用MTT法测定细胞成活率,流式细胞仪测试样品的细胞周期与凋亡率。结果在较低压力下(100mmHg)A10细胞成活率增加,细胞增殖,处于G0/G1期的A10细胞百分比显著降低,增殖指数(PI)显著升高,凋亡率与正常无差异;在200mmHg的压力下成活率降低,G0/G1期A10细胞百分比显著升高,细胞增殖受抑制,凋亡率亦显著增加。结论较低的压力可促进血管平滑肌细胞增殖,而过高的压力则可诱导细胞凋亡。
Objective To investigate the effect of resting pressure on the proliferation and apoptosis of vascular smooth muscle cells. Methods Cultured embryonic rat vascular smooth muscle cell line A10 were cultured under static pressure of 80, 100 and 200 mmHg respectively under normal atmospheric pressure. Cell survival rate, cell cycle and apoptosis rate of flow cytometry were determined by MTT assay. Results Under the low pressure (100mmHg), the survival rate of A10 cells was increased. The percentage of A10 cells in G0 / G1 phase was significantly decreased, the proliferation index (PI) was significantly increased and the apoptosis rate was no difference with A10 cells at 200mmHg Under the pressure, the survival rate decreased, the percentage of A10 cells in G0 / G1 phase increased significantly, the cell proliferation was inhibited, and the apoptosis rate was also significantly increased. Conclusions Lower pressure can promote the proliferation of vascular smooth muscle cells, while excessive pressure can induce apoptosis.
