采用形态学方法和SSR分子标记技术,对甘蓝型杂交油菜贵杂4号及其亲本进行杂种性状表现、DNA指纹图谱和种子纯度的研究结果表明:田间形态学观测结果杂种性状表现亲子间存在明显亲子关系和母本效应,利用作物的这一共性特点可对作物种子真实性和纯度进行鉴定;采用SSR分析从100对SSR引物中筛选出5对多态性很强且极易识别的特殊引物,通过这5对特殊引物的指纹组合构建了贵杂4号及其亲本的DNA指纹图谱;利用遗传决定的蛋白质组分不受环境影响和其在组分上的差异性反映的是基因型的原理,通过特殊引物组合对种子的扩增图,可快速有效的对种子真实性和纯度进行鉴定。
Morphological methods and SSR molecular markers were used to characterize the hybrid traits of Qaza 4 and its parents. The DNA fingerprinting and purity of the seeds showed that there were obvious inter-parental and hybrid phenotypes Parent-child relationship and maternal effect. The commonality of crops can be used to identify the authenticity and purity of crop seeds. Five hundred pairs of SSR primers were screened for five pairs of primers which are highly polymorphic and easily identifiable , DNA fingerprinting of Guzhao 4 and its parents was constructed by fingerprinting of these 5 pairs of special primers. The use of genetically determined protein components is unaffected by the environment and its component difference reflects the genotype Principle, through the special primer combination of seed amplification map, can quickly and effectively identify the authenticity and purity of the seed.
