目的 在分子水平上研究阿司匹林和谷胱甘肽(GSH)对酵母多糖诱导的小鼠肝组织肿瘤坏死因子-α(TNF-α)基因表达的影响。方法 分别给予小鼠低、高剂量阿司匹林(9mg、16mg·kg~(-1),ig)及GSH(20mg·kg~(-1),ip),然后给予酵母多糖(0.5g·kg~(-1),ip)。逆转录-聚合酶链反应(RT-PCR)检测肝组织TNF-αmRNA水平,放射免疫分析法检测TNF-α蛋白水平。结果 损伤组肝组织TNF-αmRNA及蛋白水平均显著高于对照组;低、高剂量阿司匹林保护组和GSH保护组肝组织TNF-αmRNA、蛋白水平都低于损伤组。结论 阿司匹林和GSH均可抑制由酵母多糖诱导的小鼠肝组织TNF-αmRNA及蛋白的表达。
Objective To investigate the effects of aspirin and glutathione (GSH) on the gene expression of tumor necrosis factor-α (TNF-α) induced by zymosan in mice liver at the molecular level. Methods Low and high dose aspirin (9 mg, 16 mg · kg -1, ig) and GSH (20 mg · kg -1, ip) were given respectively to mice. -1), ip). The levels of TNF-α mRNA in liver tissue were detected by reverse transcription-polymerase chain reaction (RT-PCR) and the level of TNF-α protein by radioimmunoassay. Results The levels of TNF-αmRNA and protein in liver tissue of injury group were significantly higher than those in control group. The levels of TNF-αmRNA and protein in liver tissue of low and high dose aspirin protection group and GSH protection group were lower than those of injury group. Conclusion Both aspirin and GSH can inhibit the expression of TNF-α mRNA and protein in the liver of mice induced by zymosan.
