应用细胞培养和间接免疫荧光法测定脐带血、成人外周血、成人骨髓的T淋巴细胞集落形成情况及培养前后淋巴细胞亚群的变化。结果表明:脐带血的CFU-TL显著低于成人外周血(P<0.01),略低于成人骨髓(P<0.05);培养前脐带血的CO+3、CO+4细胞含量均显著低于成人外周血(P<0.01),与成人骨髓相近似。脐带血的CD+8细胞含量与成人外周血和成人骨髓相似(P<0.05);培养后的CFU-TL中CD+3细胞含量脐带血与成人外周血、成人骨髓几乎相同(P>0.05),但其中CD+4细胞含量脐带血明显低于成人外周血及成人骨髓(P<0.01),而脐带血的CD+8则明显高于成人外周血(P<0.01)和成人骨髓(P<0.05),致使培养后CFU-TL中脐血CD+4/CD+8显著低于成人外周血和成人骨髓。另培养前的脐带血,成人骨髓中CD+4、CD+8细胞阳性百分率之和超过CD+3百分率,出现CD+4、CD+8双标记细胞
The formation of T lymphocytes in cord blood, adult peripheral blood and adult bone marrow and the changes of lymphocyte subsets before and after culture were measured by cell culture and indirect immunofluorescence. The results showed that the CFU-TL of umbilical cord blood was significantly lower than that of adult peripheral blood (P <0.01) and slightly lower than that of adult bone marrow (P <0.05).
