目的:研究5,7-二甲氧基黄酮(5,7-dimethoxyflavone,DMF)诱导人前单核细胞系U937细胞凋亡作用。方法:体外培养U937细胞,PI染色FCM分析Sub-G1细胞百分率;ELISA法检测细胞培养液组蛋白/DNA碎片、Caspase-9和Caspase-3活性。结果:DMF增高U937细胞Sub-G1百分率(P<0.05),呈浓度依赖性。DMF以浓度依赖方式刺激U937细胞组蛋白/DNA碎片渗漏(P<0.05)。DMF能有效诱导U937细胞Caspase-9和Caspase-3活化。结论:DMF诱导U937细胞凋亡作用与其激活线粒体途径相关。
AIM: To investigate the apoptosis of human monocytic U937 cells induced by 5,7-dimethoxyflavone (DMF). Methods: U937 cells were cultured in vitro, the percentage of Sub-G1 cells was analyzed by PI staining and the activity of Caspase-9 and Caspase-3 were detected by ELISA. Results: DMF increased the percentage of Sub-G1 in U937 cells (P <0.05) in a concentration-dependent manner. DMF stimulated histone / DNA fragment leakage in U937 cells in a concentration-dependent manner (P <0.05). DMF can effectively induce the activation of Caspase-9 and Caspase-3 in U937 cells. Conclusion: The apoptosis induced by DMF in U937 cells is related to its activation of mitochondria pathway.
