目的:探讨竹节三七总皂苷(total saponins of Panax japonicus,TSPJ)对胶原诱导型关节炎大鼠免疫功能的影响。方法:注射Ⅱ型胶原蛋白与完全弗氏佐剂的乳化剂建立大鼠关节炎模型。观察大鼠关节炎指数检测竹节三七总皂苷对关节炎模型大鼠的作用效果。采用竹节三七总皂苷治疗,检测关节炎大鼠巨噬细胞碳清除率、血清溶血素值,二硝基氟苯诱导的大鼠迟发性超敏反应(DTH),放射免疫法检测血中IL-2、TNF-α含量,苏木素-伊红染色观察脾组织病理变化,观察TSPJ对类风湿性关节炎模型大鼠作用机理。结果:与模型组比较,25 mg/kg TSPJ、50 mg/kg TSPJ、100 mg/kg TSPJ能明显减轻降低大鼠关节炎指数;对造模大鼠胸腺指数及碳清除率无明显影响;50 mg/kg TSPJ、100 mg/kg TSPJ降低血清溶血素水平;100mg/kg TSPJ能降低血中IL-2、TNF-α含量;100mg/kg TSPJ对DTH反应有抑制作用,并抑制脾组织增生。结论:竹节三七总皂苷能抑制类风湿性关节炎大鼠免疫反应,这可能是其治疗风湿类疾病的机制之一。
Objective: To investigate the effect of Total saponins of Panax japonicus (TSPJ) on immune function in rats with collagen-induced arthritis. Methods: The rat model of arthritis was established by injecting type Ⅱ collagen and complete Freund’s adjuvant. To observe the effect of rat Panax notoginseng saponins on arthritis rat model by arthritis index. Panax notoginseng total saponin was used to detect the macrophage carbon clearance, serum hemolysin, dinitrofluorobenzene-induced delayed hypersensitivity (DTH) in rats, radioimmunoassay The contents of IL-2, TNF-α, and hematoxylin-eosin staining were used to observe the pathological changes of spleen tissue. The mechanism of action of TSPJ on rheumatoid arthritis model rats was observed. Results: Compared with model group, 25 mg / kg TSPJ, 50 mg / kg TSPJ and 100 mg / kg TSPJ could obviously reduce the arthritis index of rats, but had no significant effect on thymus index and carbon clearance rate in model rats. mg / kg TSPJ, 100 mg / kg TSPJ could reduce the level of serum hemolysin; 100 mg / kg TSPJ could reduce the content of IL-2 and TNF-α in blood; 100 mg / kg TSPJ could inhibit the DTH reaction and inhibit the proliferation of spleen tissue. Conclusion: SSP can inhibit the immune response in rats with rheumatoid arthritis, which may be one of the mechanisms for the treatment of rheumatoid diseases.
