目的 :通过立体定向在 SD大鼠右尾状核区接种 C6 细胞 ,建立类似于人脑胶质瘤的动物模型。方法 :SD大鼠在立体定向条件下 ,左右尾状核区接种 1× 10 6 个 C6 胶质瘤细胞 ,接种后观察实验大鼠的生成状态 ,分别于接种后 1,2 ,3周时进行 MRI检查。在实验三周时解剖标本 ,做组织病理学和 GFAP免疫组化检查。结果 :该方法建立的胶质瘤动物模型在组织病理学上接近人脑胶质瘤 ,而且具有颅内生长稳定 ,成瘤率高 ,没有颅外种植生长 ,实验周期短 ,重复性好。结论 :该大鼠脑胶质瘤动物模型能够满足胶质瘤实验治疗研究的需要 ,而且在细胞接种后 1~ 2周 ,为实验治疗较好的时机。
OBJECTIVE: To establish an animal model similar to human glioma by stereotaxically implanting C6 cells in the right caudate nucleus of SD rats. METHODS: SD rats were inoculated with 1 × 10 6 C6 glioma cells in the left and right caudate nucleus under stereotactic conditions. The formation of experimental rats was observed after inoculation, and the rats were respectively subjected to 1, 2, 3 weeks after inoculation MRI examination. The specimens were dissected three weeks after the experiment and histopathology and GFAP immunohistochemistry were performed. Results: The animal model of glioma established by this method is histopathologically close to human glioma, and has the advantages of stable intracranial growth, high tumor formation rate, no extracranial growth, short experimental period and good repeatability. Conclusion: The animal model of glioma in rats can meet the need of experimental treatment of glioma, and the best time for experimental treatment is 1 to 2 weeks after cell inoculation.
