目的:探讨癌—睾丸抗原SSX-2基因在儿童急性淋巴细胞性白血病(ALL)外周血单个核细胞中的表达,为判断ALL预后提供理论依据。方法:采用逆转录—聚合酶链反应,对51例ALL患儿外周血单个核细胞进行SSX-2mRNA检测,并进行SSX-2基因表达与临床特征的相关性分析;以30例非肿瘤患儿为对照组。结果:ALL初治患儿SSX-2 mRNA表达率为34.78%,对照组无一例表达。ALL初治组表达率明显高于对照组(P<0.05),ALL初治组表达率高于ALL完全缓解组表达率7.14%(P<0.05)。SSX-2基因的表达与儿童ALL性别、年龄、初诊时外周血白细胞数、免疫分型以及临床危险分级无关(P>0.05)。结论:儿童ALL患者SSX-2mRNA表达率较高,SSX-2mRNA表达可能与儿童ALL的发生、发展有关;SSX-2基因检测对儿童ALL近期疗效的评估、指导临床治疗有一定意义。
Objective: To investigate the expression of cancer-testis SSX-2 gene in peripheral blood mononuclear cells of children with acute lymphoblastic leukemia (ALL), and to provide a theoretical basis for judging the prognosis of ALL. Methods: The SSX-2 mRNA of peripheral blood mononuclear cells from 51 children with ALL was detected by reverse transcription-polymerase chain reaction (RT-PCR), and the correlation between SSX-2 gene expression and clinical features was analyzed. Thirty non-tumor children For the control group. Results: The expression of SSX-2 mRNA in ALL patients was 34.78%, but none in the control group. The expression rate of ALL group was significantly higher than that of the control group (P <0.05). The expression rate of ALL group was higher than that of ALL group (7.14%, P <0.05). The expression of SSX-2 gene was not related to the gender, age, number of peripheral white blood cells, immunophenotype and clinical risk grade of ALL children (P> 0.05). Conclusion: SSX-2mRNA expression in childhood ALL patients is high, and SSX-2mRNA expression may be related to the occurrence and development of childhood ALL. The detection of SSX-2 gene in children with ALL is helpful in evaluating the clinical efficacy of ALL and guiding the clinical treatment.
