目的探讨Graves病(GD)患者糖皮质激素诱导的肿瘤坏死因子受体(GITR)mRNA表达水平的变化及意义。方法用实时荧光定量PCR法检测22例GD未治疗组、23例GD临床缓解组及18例健康对照组外周血单个核细胞(PBMC)GITR mR-NA相对表达水平;用化学发光法测定各组血浆中甲状腺激素水平,并评价GD未治疗组GITR mRNA相对表达水平与甲状腺激素水平的相关性。结果GD未治疗组GITR mRNA相对表达水平明显低于临床缓解组和健康对照组(χ2=18.48,P<0.01;χ2=18.26,P<0.01),而GD临床缓解组与健康对照组差异无统计学意义(χ2=0.12,P>0.05)。GD未治疗组GITRmRNA相对表达水平与FT3、FT4水平均呈负相关(r=-0.509、r=-0.483,P均<0.05),而与TSH呈正相关(r=0.458,P<0.05)。结论GD的发生可能与GD患者体内GITR基因表达下调有关。
Objective To investigate the changes and significance of glucocorticoid-induced tumor necrosis factor receptor (GITR) mRNA expression in Graves disease (GD). Methods The relative expression levels of GITR mR-NA in peripheral blood mononuclear cells (PBMCs) of 22 GD untreated patients, 23 GD remission patients and 18 healthy controls were detected by real-time fluorescence quantitative PCR. The levels of GITR mR- Plasma thyroid hormone levels, and evaluation of GD untreated group GITR mRNA relative expression levels and thyroid hormone levels. Results The relative expression level of GITR mRNA in GD untreated group was significantly lower than that in clinical remission group and healthy control group (χ2 = 18.48, P <0.01; χ2 = 18.26, P <0.01), but there was no statistical difference between GD remission group and healthy control group Significance (χ2 = 0.12, P> 0.05). The relative expression levels of GITR mRNA in GD untreated group were negatively correlated with FT3 and FT4 levels (r = -0.509, r = -0.483, P <0.05), but positively correlated with TSH (r = 0.458, P <0.05). Conclusion The occurrence of GD may be related to the down-regulation of GITR gene expression in GD patients.
