为研究外源性Rb 和P53 基因导入血管平滑肌细胞后对cfos 和cjun 基因表达的调控作用, 将Rb 基因或P53 基因重组腺病毒载体转染人脐动脉血管平滑肌细胞, 应用RTPCR 和免疫组化法检测cfos、cjun 的mRNA及蛋白质表达水平, 以DNA 琼脂糖凝胶电泳和β半乳糖苷酶染色分别观察细胞凋亡和细胞衰老. 结果显示, 野生型P53 基因导入可诱导平滑肌细胞凋亡,cfos 和cjun mRNA 及蛋白质表达水平显著增高. 外源性Rb 基因导入能促进细胞衰老, 下调cfos 基因表达, 但对cjun 基因表达没有明显影响
In order to study the regulatory effect of exogenous Rb and P53 gene on vascular smooth muscle cells after c fos and c jun gene expression, the Rb gene or P53 gene recombinant adenovirus vector transfected human umbilical artery smooth muscle cells, using RT PCR and immunohistochemistry c fos, c jun mRNA and protein expression levels, DNA agarose gel electrophoresis and β galactosidase staining were observed apoptosis and cell senescence. The results showed that the wild-type P53 gene can induce apoptosis of smooth muscle cells, c fos and c jun mRNA and protein expression levels were significantly higher. Exogenous Rb gene can promote cell senescence, down c fos gene expression, but c jun gene expression was not significantly affected
