目的:建立HPLC法测定珍菊降压片中绿原酸与氢氯噻嗪的含量。方法:采用反相高效液相色谱法,C18色谱柱,流动相为甲醇-0.1%磷酸溶液(20∶80),检测波长为327 nm,流速为1.0 mL.min-1,柱温40℃。结果:绿原酸进样量在0.049 6~0.496μg(r=0.999 5)、氢氯噻嗪进样量在1.002~10.02μg范围内(r=0.999 8)线性关系良好,该制剂中绿原酸9次测定的平均回收率为101.0%,RSD 2.0%;氢氯噻嗪9次测定平均回收率为100.1%,RSD 1.4%。结论:本方法简便、准确、可靠,可用于珍菊降压片中绿原酸与氢氯噻嗪的含量测定。
Objective: To establish HPLC method for the determination of chlorogenic acid and hydrochlorothiazide in Zhenju Jiangya Tablets. Methods: A RP-HPLC method was used on a C18 column. The mobile phase consisted of methanol-0.1% phosphoric acid solution (20:80). The detection wavelength was 327 nm, the flow rate was 1.0 mL.min-1, and the column temperature was 40°C. Results: The chlorogenic acid injection amount was 0.049 6~0.496 μg (r=0.999 5), and the hydrochlorothiazide injection amount was in the range of 1.002~10.02 μg (r=0.999 8). The linear relationship was good, and the chlorogenic acid was 9 times in the preparation. The average recovery determined was 101.0% with an RSD of 2.0%; the average recovery of hydrochlorothiazide 9 determinations was 100.1% and the RSD was 1.4%. Conclusion: This method is simple, accurate and reliable. It can be used for the determination of chlorogenic acid and hydrochlorothiazide in Zhenju Jiangya Tablets.
