目的探讨骨桥蛋白(osteoponin,OPN)和骨形态发生蛋白-2 bone morphogenetic protein-2,BMP-2)及其mRNA在骨肉瘤组织中的表达,分析OPN和BMP-2蛋白及其mRNA与骨肉瘤的临床分期、病理分级、浸润、转移及预后的关系。方法应用免疫组化和原位杂交方法检测105例骨肿瘤组织中的OPN、BMP-2蛋白及其mRNA的表达,并对其中85例骨肉瘤主要临床资料、病理分级及临床相关参数进行比较,用χ~2检验进行统计学处理。结果 OPN、BMP-2蛋白及其mRNA阳性表达在骨肉瘤中明显高于骨软骨瘤阳性表达(P<0.01);在骨肉瘤临床分期中Ⅱb期、Ⅲ期明显高于Ⅰ期、Ⅱa期(P<0.01),并随着病理分级恶性程度增高而显著增加(P<0.01),在软组织浸润和转移组中明显高于非浸润和非转移组(P<0.01)。结论 OPN、BMP-2蛋白及其mRNA在骨肉瘤中不同程度异常表达与肿瘤的临床分期、病理分级和浸润、转移呈正相关,提示OPN、BMP-2蛋白及其mRNA高表达在骨肉瘤的发生、发展和浸润转移过程中发挥重要作用。
Objective To investigate the expression of osteopontin (OPN) and bone morphogenetic protein-2 (BMP-2) and their mRNA in osteosarcoma tissues. The clinical stage of neoplasm, pathological grade, invasion, metastasis and prognosis. Methods The expressions of OPN and BMP-2 protein and their mRNA in 105 cases of osteosarcoma were detected by immunohistochemistry and in situ hybridization. The main clinical data, pathological grading and clinically relevant parameters of 85 cases of osteosarcoma were compared. Using χ ~ 2 test for statistical analysis. Results The positive expression of OPN and BMP-2 protein and their mRNA in osteosarcoma was significantly higher than that in osteochondroma (P <0.01). In osteosarcoma stage Ⅱb, Ⅲ was significantly higher than in stage Ⅰ, Ⅱa P <0.01), and increased with the increase of pathological grade (P <0.01), which was significantly higher in infiltration and metastasis of soft tissue than in non-infiltration and non-metastasis group (P <0.01). Conclusion The abnormal expression of OPN, BMP-2 protein and their mRNA in osteosarcoma is positively correlated with the clinical stage, pathological grade, infiltration and metastasis of tumor, which indicates that the expression of OPN, BMP-2 protein and their mRNA are highly correlated with the occurrence of osteosarcoma , Play an important role in the development and infiltration of metastasis.
