Fresia refractaKlat inflorescence explants regenerated new plants via direct somatic embryogenesis on modified N6 medium containing 2 mg / L IAA and 3 mg / LBAP. An interesting phenomenon in this morphogenesis is that all the somatic embryos occur at the lower end of the original morphology of the axon of the inflorescence (called the embryogenesis end, EE), while at the morphological upper end Non-embryogenic terminal, NEE) somatic embryogenesis. The polarity of this morphogenesis has nothing to do with the direction of gravity and the placement of explants on the medium. In the early stages of culture, initiation cell division was observed only at the embryogenic site. SDS-polyacrylamide gel electrophoresis results showed that there were two special polypeptide components in the embryogenesis end of the explants cultured for 1 day, while the two polypeptides were not detected in the non-embryogenic end. High performance liquid chromatography analysis showed that there was no significant difference in endogenous hormones (IAA) content at both ends of explant before culture. However, after a period of incubation, the content of IAA at embryogenesis was significantly higher than that at non-embryogenesis, indicating that endogenous hormones played a key role in the induction of somatic embryogenesis. During the induction of Solanum solanum embryogenesis, there is a possibility that this system may become a useful experimental material for studying the mechanism of somatic embryogenesis due to the difference in molecular level and physiological level at both ends of the axon.
