目的探讨神经母细胞瘤细胞(SH-SY5Y)和皮层神经元对神经毒物乙酸铅和硫酸铝敏感性的差异,为神经元体外替代方法及实验研究提供一定的数据支持。方法原代培养皮层神经元及培养细胞系SH-SY5Y,乙酸铅和硫酸铝染毒浓度分别为25、50、100、200、400、800、1600、3200μmol/L,作用时间分别为3和24h,测定并计算细胞存活率和半数致死浓度(IC50)值。结果除硫酸铝作用24h外,皮层神经元的IC50值均低于SH-SY5Y;SH-SY5Y对乙酸铅IC50值均高于硫酸铝;皮层神经元对乙酸铅的IC50值低于硫酸铝。结论皮层神经元对乙酸铅和硫酸铝的毒性较SH-SY5Y敏感。若以皮层神经元为靶细胞,则乙酸铅相对敏感,若以SH-SY5Y作为靶细胞,则硫酸铝则更为敏感。
Objective To investigate the difference of sensitivities of neurotoxicity lead acetate and aluminum sulfate between neuroblastoma cells (SH-SY5Y) and cortical neurons, and to provide some data support for the in vitro neuronal replacement methods and experimental studies. Methods Primary culture cortical neurons and cultured cell line SH-SY5Y, lead acetate and aluminum sulfate concentrations were 25,50,100,200,400,800,1600,3200μmol / L, the action time was 3 and 24h The cell viability and the median lethal concentration (IC50) were measured and calculated. Results The IC50 of cortical neurons was lower than that of SH-SY5Y except aluminum sulfate for 24 h. The IC50 values of SH-SY5Y for lead acetate were all higher than those of aluminum sulfate. The IC50 value of cortical neurons to lead acetate was lower than that of aluminum sulfate. Conclusion The cortical neurons are more sensitive to lead acetate and aluminum sulfate than SH-SY5Y. Lead acetate is relatively sensitive if cortical neurons are target cells, and aluminum sulfate is more sensitive if SH-SY5Y is used as a target cell.
