为鉴定引起2010年江西省急性出血性结膜炎(Acute hemorrhagic conjunctivitis,AHC)疫情的病原体,并对其进行基因进化分析,本研究采集了20份门诊AHC患者眼结膜拭子,对其进行病毒分离,随后通过逆转录-聚合酶链式反应(Reverse transcription-polymerase chain reaction,RT-PCR)方法分别检测阳性分离物中肠道病毒70型(Human enterovirus type 70,EV70)、柯萨奇病毒A24变异株(Coxsackievirus A24variant,CV-A24v)和腺病毒。并对CV-A24v分离株进行VP1区和3C区全序列测定,分析其与全球流行CV-A24v的基因进化关系。20份标本中有10份病毒分离阳性,PCR检测证明引起本次AHC流行的病原体为CV-A24v。基于3C区构建的基因进化树表明10株江西CV-A24v与全球2010年后分离到的其它CV-A24v同属于GenotypeⅣ基因型的Cluster 5群(GⅣ-C5),而且在GⅣ-C5内江西CV-A24v又分属于A和B两个传播链。基于VP1区构建的基因进化树将全球2000年后分离到的CV-A24v分成5组Group1~5(G1~5),江西CV-A24v属于G5,这些毒株在VP1区进化树中同样分属于A和B两个传播链。本研究表明2010年至少有两个CV-A24v传播链在江西同时流行。VP1区基因进化树对全球2000年后分离到的CV-A24v的组群划分与3C区基本一致,而且与3C区呈现同样显著的年代聚集性。
To identify the pathogens causing acute hemorrhagic conjunctivitis (ICC) outbreak in Jiangxi province in 2010 and to analyze the gene evolution, 20 conjunctival swabs from outpatients with AHC were collected and subjected to virus isolation , And then the positive isolates were detected by RT-PCR (reverse transcription-polymerase chain reaction, RT-PCR) Coxsackievirus A24variant (CV-A24v) and adenovirus. The full-length VP1 and 3C regions of CV-A24v isolates were assayed and their phylogenetic relationships with the global epidemic CV-A24v were analyzed. Of the 20 specimens, 10 were positive for virus isolation, and the PCR test showed that the pathogen causing the AHC epidemic was CV-A24v. The phylogenetic tree based on 3C showed that 10 CV-A24v isolates in Jiangxi province and other CV-A24v isolates from the world after 2010 belong to the same GenotypeⅣ genotype Cluster 5 (GⅣ-C5) -A24v is divided into A and B two transmission chain. Based on the phylogenetic tree constructed based on the VP1 region, the CV-A24v isolated from the world after 2000 was divided into five groups of Group1-5 (G1-5), and the Jiangxi CV-A24v belonged to G5. These strains belonged equally to A and B two transmission chain. This study shows that at least two CV-A24v transmission lines in 2010 were endemic in Jiangxi at the same time. VP1 region phylogenetic tree of the global cluster of CV-A24v isolated after 2000 and the 3C area is basically the same, and with the 3C area showed the same significant age aggregation.
