探讨肉桂酸对人肺腺癌A5 49细胞诱导分化作用及可能机制。方法 应用 2mmol/L终浓度肉桂酸作用于人肺腺癌A5 49细胞 ,观察体外生长增殖情况、双层软琼脂集落形成率并用流式细胞仪测定细胞周期及p5 3蛋白表达。结果 细胞体外生长速度减慢 ;双层软琼脂集落形成减少 ;细胞DNA合成能力降低 ;细胞周期出现向G1/G0期移行的特征性动力学改变 ;p5 3蛋白表达减少。结论 肉桂酸通过激活人肺腺癌细胞分化相基因表达启动细胞分化并达到恶性逆转 ,是一种有应用潜力的诱导分化剂。
To investigate the effect of cinnamic acid on the differentiation of human lung adenocarcinoma A5 49 cells and its possible mechanism. Methods Cinnamic acid at a final concentration of 2 mmol/L was used to treat human lung adenocarcinoma A549 cells. Growth and proliferation in vitro, double soft agar colony formation rate, and cell cycle and p53 protein expression were measured by flow cytometry. Results The growth rate of the cells in vitro was slowed down; the formation of double soft agar colonies was reduced; the ability of DNA synthesis was decreased; the characteristic dynamics of the cell cycle to the G1/G0 transition; the expression of p53 protein was decreased. Conclusion Cinnamic acid induces cell differentiation and achieves malignant reversal by activating the differentiation gene expression of human lung adenocarcinoma cells. It is an inducing agent with potential for application.
