以抗坏血酸-Fe~(3+)作为自由基生成体系,结合琼脂糖电泳和激光密度扫描等方法,观察了自由基对质粒pBR322 DNA的损伤及过氧化氢酶和超氧化物歧化酶的防护效能。结果表明,该体系生成的羟自由基可造成DNA的单链断裂而使pBR322的超螺旋构型转变为开环构型;过氧化氢酶对羟自由基所致DNA损伤的抑制作用显著,而超氧化物歧化酶无明显抑制作用。
Ascorbic acid-Fe ~ (3+) was used as a free-radical generating system. The effects of free radicals on DNA damage of pBR322 and the protective efficacy of catalase and superoxide dismutase were observed by agarose gel electrophoresis and laser scanning densitometry. . The results showed that the hydroxyl radical generated by this system could cause the single-strand breaks of DNA and the trans-helix configuration of pBR322 to open-loop configuration. The catalase had a significant inhibitory effect on the DNA damage induced by hydroxyl radicals Superoxide dismutase no significant inhibitory effect.
