自青海北山林场桦树根围分离筛选到芽胞杆菌菌株166株,根据菌落特征分为6个类群,选择其中31株菌株纯化保存并进行BOX-PCR及ERIC-PCR指纹图谱分析、16S rDNA及gyrB基因序列分析,鉴定出球形赖氨酸芽胞杆菌Lysinibacillus sphaericus 12株、解淀粉芽胞杆菌Bacillus amyloliq-uefaciens 5株、韦氏芽胞杆菌Bacillus weihenstephanensis 7株、芽胞杆菌Bacillus sp.5株、苏云金芽胞杆菌Bacillus thuringiensis 2株。通过平板对峙试验筛选得到11株对油菜菌核病原真菌Sclerotiniasclerotiorum具有显著拮抗活性的菌株。以MALDI-TOF-MS质谱分析拮抗菌株QB11的活性成分,可产生脂肽化合物表面活性素和泛革素,表明其拮抗活性与脂肽化合物的合成和分泌有关。
A total of 166 strains of Bacillus were isolated from the bark of birch in Beishan Forest Farm of Qinghai Province and classified into 6 groups according to the characteristics of colonies. 31 strains were selected and purified, and analyzed by the methods of BOX-PCR and ERIC-PCR, 16S rDNA and gyrB Gene sequence analysis identified 12 strains of Lysinibacillus sphaericus, 5 strains of Bacillus amyloliq-uefaciens, 7 strains of Bacillus weihenstephanensis, 5 strains of Bacillus sp.5 and 3 strains of Bacillus thuringiensis 2 strains. Eleven strains with significant antagonistic activity against Sclerotinasclerotiorum were identified by slab confrontation test. Analysis of the active ingredient of the antagonistic strain QB11 by MALDI-TOF-MS mass spectrometry yielded the lipopeptide compound surfactant and pan-tannin, indicating that its antagonistic activity is related to the synthesis and secretion of lipopeptide compounds.
