目的通过观察不同浓度胰岛素对视网膜M櫣ller细胞内谷氨酸(glutamate,Glu)-谷氨酰胺循环(glutamine,Gln)及谷胱甘肽(glutathione,GSH)的影响,探讨其在视网膜神经损伤中的作用。方法利用生物化学检测方法,观察并测定体外培养的兔视网膜Müller细胞内Glu、Gln合成酶(glutamine synthetase,GS)、Gln及GSH在不同胰岛素条件下的含量变化。结果随着胰岛素浓度的升高及作用时间的延长,Müller细胞内Glu、Gln、GS和GSH的含量均呈下降趋势,且与对照组相比明显降低(P<0.01)。结论高浓度胰岛素能够减少视网膜M櫣ller细胞内Glu的含量,降低GS活性,减少Gln、GSH合成,从而减弱其保护神经元的功能。
Objective To investigate the effects of different concentrations of insulin on glutamate (Gln) and glutathione (GSH) in retinal Müller cells and to explore the role of insulin in retinal nerve injury In the role. Methods The biochemical assay was used to observe and measure the contents of Glu, Gln synthetase (GS), Gln and GSH in cultured retinal Müller cells cultured in vitro under different insulin conditions. Results The contents of Glu, Gln, GS and GSH in Müller cells decreased with the increase of insulin concentration and the prolongation of action time, and were significantly decreased compared with the control group (P <0.01). Conclusion High concentration of insulin can reduce the content of Glu in retinal Müller cells, decrease the activity of GS, decrease the synthesis of Gln and GSH, and weaken the function of protecting neurons.
