目的观察肥胖大鼠肝脏和骨骼肌叉头状因子O1(FoxO1)的表达,探讨FoxO1在肥胖和胰岛素抵抗发生中的作用。方法 30只SD大鼠随机分为对照组(NC,常规饲料)和高脂组(HF,高脂饲料)。饲养10周后测定有关指标。结果 10周后与NC组相比,HF组体重、血糖、胰岛素、TG、TC、腹内脂肪/体重均升高,肝脏和骨骼肌FoxO1mRNA表达量分别增加84%和88%(P<0.01)。TG、HO-MA-IR及腹内脂肪/体重是肝脏FoxO1mRNA表达的独立相关因素,HOMA-IR及腹内脂肪/体重是骨骼肌FoxO1表达水平的主要影响因素。结论肥胖大鼠肝脏和骨骼肌FoxO1表达量升高,可能是肥胖状态下引发胰岛素抵抗的机制之一。
Objective To observe the expression of FoxO1 in liver and skeletal muscle of obese rats and to explore the role of FoxO1 in the pathogenesis of obesity and insulin resistance. Methods Thirty SD rats were randomly divided into control group (NC, normal diet) and high fat diet (HF, high fat diet). After 10 weeks of feeding, the relevant indexes were measured. Results After 10 weeks, the body weight, blood glucose, insulin, TG, TC and intra-abdominal fat / body weight of HF group were increased compared with NC group. The expression of FoxO1 mRNA in liver and skeletal muscle increased by 84% and 88% . TG, HO-MA-IR and intra-abdominal fat / body weight were the independent relevant factors of liver FoxO1 mRNA expression. HOMA-IR and intra-abdominal fat / body weight were the main influencing factors of skeletal muscle FoxO1 expression. Conclusion The increased FoxO1 expression in the liver and skeletal muscle of obese rats may be one of the mechanisms that lead to insulin resistance in obesity.
