研究了维多利亚蓝B与利血平的显色反应,建立了测定利血平的高灵敏分光光度法。在弱碱性条件下,利血平的水解产物和维多利亚蓝B形成具有正、负吸收峰的蓝色离子缔合物,最大正吸收波长位于612 nm,最大负吸收波长位于506 nm,线性范围为0~5.0μg/mL(正吸收)和0~4.5μg/mL(负吸收),表观摩尔吸光系数(ε)分别为2.26×105L.mol-1.cm-1(正吸收)和9.09×104L.mol-1.cm-1(负吸收)。当用双波长叠加法时,ε值为3.20×105L.mol-1.cm-1。探讨了适宜的反应条件、主要分析化学性质及方法的精密度和可靠性,并用正吸收法对市售利血平注射液中利血平含量进行了测定,结果满意。
The color reaction of Victoria Blue B with reserpine was studied and a sensitive spectrophotometric method for the determination of reserpine was established. Under mildly alkaline conditions, the hydrolysates of reserpine and Victoria blue B formed blue ion complexes with positive and negative absorption peaks. The maximum positive absorption wavelength was at 612 nm and the maximum negative absorption wavelength was at 506 nm. The linear range (Positive absorption) and 0-4.5 μg / mL (negative absorption), the apparent molar absorptivities (ε) were 2.26 × 105L.mol-1.cm-1 × 104L.mol-1.cm-1 (negative absorption). When using the double wavelength superposition method, the ε value is 3.20 × 10 5 L · mol -1 · cm -1. The suitable reaction conditions, the main analytical chemistry and the precision and reliability of the method were discussed. The content of reserpine in the commercial reserpine injection was determined by the positive absorption method. The results were satisfactory.
