大豆花器小,花瓣中蛋白含量低,花瓣采集是大豆花型发育蛋白质组学研究首先要解决的难题。大豆开花期一般持续大约为30d,花瓣采集不仅需要选择恰当的时期、掌握正确的方法,还需要一定的实践经验和技巧。本研究确定了不同发育阶段的大豆花瓣采集标准;明确了采集用于蛋白质组双向电泳分析的不同发育阶段大豆花瓣所需的工作量;建立了1mm花苞大豆花瓣的采集方法。为大豆花型发育蛋白质组双向电泳分析提供了有效的样品采集方法。
Soybean flower is small, the petal protein content is low, petal collection is the first flower type development proteomics study to solve the problem. Soybean flowering generally lasts about 30d, petal collection not only need to choose the right period, the right way to master, but also need some practical experience and skills. In this study, soybean petal collection criteria were determined at different developmental stages. The workload of soybean petals collected at different developmental stages for proteome analysis was determined. A collection method of 1mm petal of soybean buds was established. It provided an effective method for sample collection for two-dimensional electrophoresis analysis of soybean flower type proteome.
