目的观察辛伐他汀对动脉粥样硬化大鼠高迁移率族蛋白1(HMGB-1)的蛋白及基因表达及斑块形态学的影响,以揭示辛伐他汀抑制炎症反应,对动脉粥样硬化的保护作用机制。方法30只大鼠随机分为3组:空白对照组、动脉粥样硬化组和辛伐他汀组,辛伐他汀灌胃进行干预,给药4周。免疫组织化学染色法观察大鼠主动脉血管动脉粥样硬化斑块内HMGB-1的蛋白表达情况,实时聚合酶链反应法检测大鼠主动脉组织HMGB-1的基因表达变化,酶联免疫吸附试验检测大鼠血清HMGB-1蛋白表达。结果(1)HMGB-1在动脉粥样硬化斑块内的表达呈强阳性,辛伐他汀组HMGB-1表达较单纯动脉粥样硬化组减少。(2)与空白对照组比较,在动脉粥样硬化斑块内动脉粥样硬化组HMGB-1基因表达水平明显升高,差异有显著性(P<0.01);辛伐他汀干预组HMGB-1基因表达低于动脉粥样硬化组(P<0.01),高于空白对照组(P<0.01)。(3)与空白对照组比较,动脉粥样硬化组及辛伐他汀组血清HMGB-1蛋白表达水平均明显升高,差异有显著性(P<0.01),辛伐他汀组血清HMGB-1表达低于动脉粥样硬化组(P<0.05)。结论(1)动脉粥样硬化大鼠主动脉血管HMGB-1的蛋白及基因表达均明显增强。(2)辛伐他汀能够减轻动脉粥样硬化大鼠的斑块形成,降低HMGB-1的蛋白及基因表达,通过减轻炎症反应的途径起到保护作用。

Results (1) The expression of HMGB-1 in atherosclerotic plaque was strongly positive. The expression of HMGB-1 in simvastatin group was lower than that in atherosclerotic group. (2) Compared with the blank control group, the expression of HMGB-1 gene in atherosclerotic plaques of atherosclerotic plaques was significantly increased (P <0.01), the levels of HMGB-1 in simvastatin intervention group The gene expression was lower than that of atherosclerosis group (P <0.01), higher than that of the blank control group (P <0.01). (3) Compared with the blank control group, the serum HMGB-1 protein expression levels in atherosclerotic and simvastatin groups were significantly increased (P <0.01), the serum HMGB-1 expression in simvastatin group Lower than atherosclerosis group (P <0.05). Conclusions (1) The protein and gene expression of HMGB-1 in the aorta of atherosclerotic rats were significantly increased. (2) Simvastatin can reduce the plaque formation in atherosclerotic rats and decrease the protein and gene expression of HMGB-1, which may play a protective role by reducing the inflammatory response.
