应用细胞封接式膜片钳方法,在Wistar大鼠的单个心室肌细胞上记录了T型、L到和B到钙通道的单通过活动.人参三醇组皂甙单体Rh160μmol/L显著抑制3种类型钙通道的活动,使其开放概率减少与开放时间缩短,对通过通道的离子流幅度无明显影响.用电子自旋共振法(ESR)测定了培养心肌细胞的自由基含量.Rh130μg/ml能显著抑制黄嘌呤(0.42mmol/L)-黄嘌呤氧化酶(5.3nmol/L)31起的自由基含量增多.
Single-pass activities of T-type, L-to, and B-to-calcium channels were recorded on single ventricular myocytes of Wistar rats using the cell seal patch clamp method. The ginsenotriol saponin monomer Rh160μmol/L significantly inhibited the activities of the three types of calcium channels, reduced the opening probability, shortened the opening time, and had no significant effect on the amplitude of the ion flow through the channel. The free radical content of cultured cardiomyocytes was determined by electron spin resonance (ESR). Rh130μg/ml can significantly inhibit the increase of free radicals content of xanthine (0.42mmol/L)-xanthine oxidase (5.3nmol/L) 31 times.
