目的:探讨二烯丙基三硫(diallytrisulfide,DATS)诱导人胃癌细胞系MGC-803凋亡与细胞内活性氧(re-activeoxygenspecies,ROS)之间的关系。方法:用12mg/L的DATS作用于体外培养的MGC-803细胞24h后,用流式细胞术检测凋亡率及细胞内活性氧水平。结果:12mg/LDATS作用于MGC-803细胞24h后,细胞凋亡率明显上升;细胞内活性氧水平与DATS呈浓度依赖性升高。用抗氧化剂NAC预处理细胞,能抑制细胞内活性氧水平的升高及DATS诱导的细胞凋亡。结论:DATS诱导胃癌细胞凋亡与细胞内活性氧产生增加有关。
Objective: To investigate the relationship between apoptosis of human gastric cancer cell line MGC-803 induced by diallytrisulfide (DATS) and intracellular reactive oxygen species (ROS). Methods: After treated with 12 mg/L DATS for 24 h in vitro, apoptosis rate and intracellular reactive oxygen species were detected by flow cytometry. RESULTS: After 24 h of treatment with 12 mg/LDATS in MGC-803 cells, the apoptotic rate increased significantly; the level of reactive oxygen species and DATS in the cells increased in a concentration-dependent manner. Pretreatment of cells with the antioxidant NAC can inhibit the increase of intracellular reactive oxygen species and DATS-induced apoptosis. Conclusion: The apoptosis of gastric cancer cells induced by DATS is related to the increase of intracellular reactive oxygen species.
