目的 :研究卡介苗 (BCG)能否诱导人单核白细胞系细胞THP 1、IL 1 2P40基因表达 ,并鉴定其胞壁蛋白的活性组份。方法 :应用逆转录聚合酶链反应 (RT PCR)法检测THP 1细胞IL 1 2P40mRNA的表达 ;采用超声破碎细胞、蔗糖密度梯度离心、SephadexG 1 5 0柱层析等方法分离卡介苗胞壁蛋白质组份。 结果 :卡介苗胞壁蛋白SephadexG 1 5 0柱层析所得组分中 ,分子量范围约 2 1～ 2 9kD的组份诱导THP 1细胞IL 1 2P40mR NA的表达增强。结论 :结果显示BCG胞壁蛋白可刺激白细胞IL 1 2P40基因的表达。
Objective: To investigate whether BCG can induce the expression of THP 1 and IL 1 2P40 genes in human monocytic leukemia cells and identify the active components of their cell wall proteins. Methods: The expression of IL-12P40 mRNA in THP1 cells was detected by reverse transcriptase-polymerase chain reaction (RT PCR). The protein components of BCG cell wall were separated by sonication, sucrose density gradient centrifugation and SephadexG 150 column chromatography . Results: The components of BCG cell wall protein SephadexG 150 column showed an increase in the expression of IL-12P40 mRNA in THP1 cells in the range of 21 ~ 29kD. Conclusion: The results show that BCG cell wall protein can stimulate leukocyte IL-12P40 gene expression.
