目的:探讨补阳还五汤对大鼠脑缺血再灌注后海马CA1区缝隙连接小体connexin43(Cx43)的影响。方法:采用颈内动脉线栓法建立大脑中动脉栓塞模型,脑缺血2 h再灌注1、3和7 d后,免疫组织荧光染色观察海马CA1区Cx43的表达情况,并通过星形胶质细胞标记物胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)的免疫染色比较星形胶质细胞的激活程度。结果:补阳还五汤组脑缺血再灌注1 d后海马CA1区Cx43的表达较模型组显著降低(P<0.05),而第3天和第7天海马CA1区Cx43的表达较模型组高(P<0.05)。同时,补阳还五汤组缺血再灌注1 d后星形胶质细胞的激活较模型组受到抑制;第3天和第7天星形胶质细胞激活均较第1天明显减弱,而且第7天时两组无明显差异。结论:补阳还五汤可能通过调控星形胶质细胞Cx43表达对脑缺血再灌注后不同时间点的损伤和修复进行调控。
Objective: To investigate the effects of Buyang Huanwu decoction on connexin43 (Cx43) in hippocampal CA1 area after cerebral ischemia-reperfusion in rats. Methods: The middle cerebral artery occlusion model was established by the method of internal carotid artery occlusion. The expression of Cx43 in the hippocampal CA1 region was observed by immunohistochemical staining at 1, 3 and 7 days after reperfusion for 2 h after cerebral ischemia. Immunostaining of glial fibrillary acidic protein (GFAP), a cellular marker, compared the activation of astrocytes. Results: The expression of Cx43 in CA1 area of hippocampus in BUYH group was significantly lower than that in model group (P <0.05), and the expression of Cx43 in hippocampal CA1 area on day 3 and 7 was significantly lower than that in model group High (P <0.05). At the same time, the activation of astrocytes in Buyanghuanwu decoction group for 1 d after ischemia-reperfusion was inhibited as compared with the model group; the activation of astrocytes on the 3rd and 7th days were significantly weaker than that on the 1st day There was no significant difference between the two groups on the 7th day. Conclusion: Buyang Huanwu decoction may regulate the injury and repair of cerebral ischemia reperfusion by regulating the expression of Cx43 in astrocytes.
