建立链脲佐菌素联合高脂喂养诱导的2型糖尿病(DM)大鼠模型并进行腹腔内葡萄糖耐量试验,评价β细胞功能,RT-PCR和Western印迹结果显示2型DM模型大鼠胰岛11β-类固醇脱氢酶1型(11β-HSD1)表达明显升高,与空腹血糖水平呈正相关;与空腹胰岛素、胰岛素和血糖曲线下面积比值呈负相关;且11β-HSD1表达与Pdx1表达负相关。提示2型DM模型大鼠11β-HSD1在胰岛表达增加,扩大局部组织糖皮质激素效应,使B细胞功能受损。
The model of type 2 diabetes mellitus (DM) induced by streptozotocin combined with high fat diet was established and the intraperitoneal glucose tolerance test was performed to evaluate the function of β cells. The results of RT-PCR and Western blot showed that 11β --His steroid dehydrogenase type 1 (11β-HSD1) expression was significantly increased, and fasting blood glucose levels were positively correlated with fasting insulin, insulin and blood glucose area under the ratio was negatively correlated; and 11β-HSD1 expression and Pdx1 negatively correlated. These results suggest that the expression of 11β-HSD1 in type 2 DM rats increases in pancreatic islets and enlarges the glucocorticoid effect in local tissues, impairs B cell function.
