目的观察重组人白细胞介素-11(rhIL-11)和重组人粒细胞集落刺激因子(rhG-CSF)联用对60Coγ射线照射所致急性放射病的疗效。方法16只比格狗随机分为对照组(5只)、对症治疗组(5只)和细胞因子治疗组(6只)。所有动物均接受4.5Gy60Coγ射线照射,并在照后当天开始治疗,对症治疗组仅给予积极的对症支持治疗,细胞因子治疗组在对症治疗的基础上,加用rhIL-11和rhG-CSF,对照组不给予任何治疗。观察动物的一般情况、外周血象、外周血有核细胞早期凋亡率和坏死率、血浆中细胞因子含量及组织病理学变化。结果细胞因子治疗组动物的45d存活率升高,外周血象也明显改善;外周血有核细胞早期凋亡率和坏死率明显降低;外周血血浆中IL-2和IFN-γ含量在早期均升高,随后IL-2迅速下降,IFN-γ含量相对比较稳定;骨髓组织病理学结果显示造血功能明显恢复。结论rhIL-11和rhG-CSF联用可促进急性放射病比格狗的造血和免疫功能恢复,是治疗急性放射病的有效方法。
Objective To observe the effect of recombinant human interleukin-11 (rhIL-11) combined with recombinant human granulocyte-colony stimulating factor (rhG-CSF) on acute radiation sickness caused by 60Coγ ray irradiation. Methods Sixteen Beagle dogs were randomly divided into control group (n = 5), symptomatic treatment group (n = 5) and cytokine treatment group (n = 6). All animals received 4.5Gy60Coγ ray irradiation, and the day after the start of treatment, symptomatic treatment group was given only symptomatic symptomatic treatment of symptomatic treatment, cytokine treatment group based on the addition of rhIL-11 and rhG-CSF, control The group did not receive any treatment. The animals were observed general situation, peripheral blood, peripheral blood mononuclear cells in early apoptosis and necrosis rates, plasma levels of cytokines and histopathological changes. Results The survival rate of 45 days in the cytokine treated group increased significantly and the peripheral blood was also significantly improved. The early apoptotic rate and necrosis rate of the peripheral blood mononuclear cells were significantly decreased. The levels of IL-2 and IFN-γ in peripheral blood plasma were increased in the early stage High, followed by rapid decline in IL-2, IFN-γ content is relatively stable; bone marrow histopathology showed hematopoietic function recovered significantly. Conclusion The combination of rhIL-11 and rhG-CSF can promote hematopoietic and immune recovery of acute radiation sickness beagle dogs, which is an effective method for the treatment of acute radiation sickness.
