目的:优选连翘叶中连翘酯苷的最佳反相硅胶(RP-18)中压制备工艺条件。方法:采用单因素多水平试验的方法,考察上样量(连翘酯苷粗品/RP-18硅胶)、洗脱流速、柱床径高比等因素时连翘酯苷得量的影响,优选出最佳纯化工艺条件。结果:最佳工艺为上样量(连翘酯苷粗品/RP-18硅胶)为1:50、洗脱流速为1.5 BV·h~(-1)、柱床径高比为1:10。结论:优选得到的连翘酯苷的最佳反相(RP-18)中压制备工艺简单、稳定、可行。
OBJECTIVE: To optimize medium-pressure preparation conditions of Forsythiaside in Forsythia suspensa leaf by reversed-phase silica gel (RP-18). Methods: The effect of forsythiaside on the sample volume (forsythiaside crude / RP-18 silica gel), elution flow rate, bed diameter-height ratio and other factors was investigated by single factor and multi-level test. The best purification process conditions. Results: The optimum conditions were as follows: the amount of sample (forsythiaside crude / RP-18 silica gel) was 1:50, the elution flow rate was 1.5 BV · h ~ (-1), and the bed height-diameter ratio was 1:10. CONCLUSION: The optimal reversed phase (RP-18) medium pressure preparation process of the optimal Forsythiaside is simple, stable and feasible.
