目的观察KGFR腺病毒表达载体在脂多糖(LPS)致A549细胞损伤模型中转基因表达KGFR的情况及其促使A549细胞对抗LPS损伤的效果,为基因治疗急性肺损伤探索有效途径。方法KGFR腺病毒表达载体感染LPS致伤前后的A549细胞,通过形态学观察转基因作用对LPS致伤前后A549细胞的影响,并运用western blot检测KGFR腺病毒表达载体在LPS致伤前后的A549细胞中转基因表达KGFR蛋白情况。结果KGFR腺病毒表达载体感染LPS致伤前后的A549细胞,转基因组对抗LPS致伤作用明显,western blot检测表明KGFR腺病毒表达载体转基因表达KGFR蛋白效果显著(P<0.05)。结论KGFR腺病毒表达载体能够转基因表达KGFR蛋白,并具备转基因治疗急性肺损伤的能力。
Objective To observe the effect of KGFR adenovirus expression vector on gene expression of KGFR in A549 cell injury induced by lipopolysaccharide (LPS) and its effect on LPS-induced injury in A549 cells, and to explore an effective approach for gene therapy of acute lung injury. Methods The A549 cells were infected with KGFR adenovirus vector before and after LPS injury. The morphological changes of A549 cells treated with LPS were observed. The expression of KGFR adenovirus was detected by western blot in A549 cells before and after LPS injury Transgenic expression of KGFR protein. Results The KGFR adenovirus expression vector infected A549 cells before and after LPS injury. The transgene group had obvious anti-LPS injury effect. The results of western blot showed that KGFR adenovirus expression vector transfected with KGFR protein had a significant effect (P <0.05). Conclusion KGFR adenovirus expression vector can genetically express KGFR protein and possess the ability of transgenic treatment of acute lung injury.
