目的:本实验旨在通过分析旋复代赭汤及其拆方对反流性食管炎(RE)模型大鼠食管组织线粒体UCP2表达的影响,探讨气机升降与线粒体能量代谢调控机制的相关性,以期进一步阐释RE的病机及旋复代赭汤治疗RE的作用机理。方法:将150只健康雄性Wister大鼠随机分为10组,即对照组(正常组、模型组)、旋复代赭汤全方组及拆方组(苦降组、甘升组、升降相因组、全方去苦降组、全方去甘升组、全方去升降相因组)与西药组,每组15只。采用“4.2mm幽门夹+胃底2/3结扎术”制备酸碱混合反流性食管炎大鼠模型。术后第8天开始,旋复代赭汤全方组及拆方各组分别给予相应药液灌胃,西药组给予兰索拉唑+莫沙必利灌胃,对照组(正常组+模型组)均给予等量生理盐水灌胃。干预14天后,第22天处死所有模型大鼠。在光镜下观察食管黏膜组织病理变化,采用Western-blot法检测各组RE模型大鼠食管组织线粒体中UCP2的相对表达量。结果:通过Western-blot相对灰度的比较分析,正常组UCP2的相对表达量最低,同全方组及西药组比较无明显差异(P>0.05);而全方组及西药组UCP2的相对表达量明显低于模型组(P<0.01);全方去苦降组、甘升组、升降相因组与模型组比较,结果有差异(P<0.05);其余各组(全方去甘升组、全方去升降相因组、苦降组)UCP2的相对表达量与模型组比较无明显差异(P>0.05),不具有统计学意义。结论:旋复代赭汤可以明显降低RE大鼠食管组织线粒体UCP2的表达量,表明此方能够很好的调节RE大鼠食管线粒体能量代谢,以此推断该方能明显改善RE大鼠食管下端括约肌松弛,在缓解胃食管反流方面具有良好的疗效。
Objective: The purpose of this experiment is to analyze the influence of Xuan Fu Daizhe Decoction and its decomposed formulas on the expression of UCP2 in esophageal tissue of rats with reflux esophagitis (RE) In order to further explain the pathogenesis of RE and the mechanism of Xuan Fu Daizhe Decoction in treating RE. Methods: One hundred and fifty healthy Wister rats were randomly divided into 10 groups, namely control group (normal group, model group), Xuanfu Daizhe decoction group and dismantling group (bitter decoction group, Ganxue decoction group, All to bitter down group, all to Gan or group, all to ascend phase group) and Western medicine group, 15 in each group. After 14 days of intervention, all model rats were sacrificed on the 22nd day. The pathological changes of esophageal mucosa were observed under light microscope. The relative expression of UCP2 in esophageal tissue mitochondria of RE model rats was detected by Western-blot. Results: The relative expression level of UCP2 in the normal group was the lowest, compared with the whole group and western medicine group (P> 0.05) by Western-blot relative gray value. The relative expression of UCP2 in the whole group and western medicine group (P <0.01). There was a significant difference between the model group and the other groups (P <0.01). The other groups There was no significant difference in the relative expression level of UCP2 between the model group and the untreated group (P> 0.05), which was not statistically significant. Conclusion: Xuanfu Daizhe Decoction can significantly reduce the expression of mitochondrial UCP2 in esophagus of RE rats, indicating that this prescription can well regulate the energy metabolism of esophageal mitochondria in RE rats, and concludes that this prescription can significantly improve the lower esophageal sphincter of RE rats Relaxation, in relieving gastroesophageal reflux has a good effect.
