目的建立测定榆黄喷雾剂中缩合鞣质含量的方法。方法供试品溶液中依次加入3%香草醛甲醇溶液、30%浓硫酸甲醇溶液各5 mL后避光显色20 min,用紫外分光光度法在500 nm波长处测定吸光度并计算样品中缩合鞣质含量。结果儿茶素对照品质量浓度在0.03~0.3 g/L范围内与吸光度呈良好线性关系,回归方程为Y=3.065 2X+0.025 7,r=0.999 3(n=10);加样回收率为99.62%,RSD为1.76%(n=6)。结论香草醛硫酸法测定缩合鞣质含量,方法简便、快速、精密度、稳定性、重复性、专属性好,适合作为榆黄喷雾剂中缩合鞣质的含量检测方法。
Objective To establish a method for the determination of condensed tannin in Yuhuang Spray. Methods 3% vanillin methanol solution and 5% 30% concentrated sulfuric acid methanol solution were respectively added into the test solution for 15 minutes and then exposed to light for 20 minutes. The absorbance was measured by ultraviolet spectrophotometry at a wavelength of 500 nm and the condensed tannage Quality content. Results The concentration of catechin reference substance in the range of 0.03 ~ 0.3 g / L showed a good linear relationship with absorbance. The regression equation was Y = 3.065 2X + 0.025 7, r = 0.999 3 (n = 10) 99.62%, RSD 1.76% (n = 6). Conclusion Vanillin sulfate method for the determination of condensed tannin content is simple, rapid, precise, stable, reproducible and specific. It is suitable for the detection of condensed tannins in elm spray.
