目的研究亚慢性砷中毒对大鼠睾丸生精细胞半胱氨酸天冬氨酸酶-3(caspase-3)表达及血清黄体生成素(LH)的影响。方法将40只健康成年清洁级SD雄性大鼠随机分为4组,分别为对照组(蒸馏水)组和0.4、2、10 mg/kg亚砷酸钠染毒组,每组10只,采用自由饮水方式进行染毒,连续染毒14周。测定大鼠血清LH浓度及生精细胞caspase-3阳性率。结果与对照组比较,各剂量亚砷酸钠染毒组大鼠生精细胞caspase-3阳性率均较高,2、10 mg/kg亚砷酸钠染毒组大鼠血清LH浓度均较低,差异有统计学意义(P<0.01,P<0.05);且随着亚砷酸钠暴露剂量的升高,大鼠生精细胞caspase-3阳性率呈上升趋势,血清LH浓度呈下降趋势。结论砷暴露导致的生精细胞caspase-3表达增加可能是其雄性生殖毒性效应的机制之一。
Objective To investigate the effect of subchronic arsenic poisoning on the expression of saspase caspase-3 and serum luteinizing hormone (LH) in rat testis. Methods Forty healthy male SD rats were randomly divided into 4 groups: control group (distilled water) and 0.4, 2, 10 mg / kg sodium arsenite exposure group, with 10 rats in each group. Drinking water for exposure, continuous exposure for 14 weeks. The concentration of serum LH and the positive rate of spermatogenic cells caspase-3 were determined. Results Compared with the control group, the positive rates of caspase-3 in spermatogenic cells of rats treated with sodium arsenite at various doses were higher than those in the control group. The concentrations of serum LH in 2,10 mg / kg sodium arsenite-treated rats were lower (P <0.01, P <0.05). The positive rate of caspase-3 in rat spermatogenic cells increased with the dose of sodium arsenite increased, and the serum LH concentration showed a decreasing trend. Conclusion The increased expression of caspase-3 in spermatogenic cells induced by arsenic may be one of the mechanisms of male reproductive toxicity.
