采用放射受体结合法及分子筛HPLC分析研究人白血病细胞株HL-60细胞核内的维甲酸(RA)受体。发现HL-60细胞核内存在RA特异结合部位。Scatchard分析表明结合呈单位点,ku为2.03nmol/L,Bmax为1352.6pmol/mg protein。分子筛HPLC分析表明结合位点的分子量约为50ku。这些结果提示HL-60细胞核内存在的RA特异结合位点具有受体性质。细胞总RNA与维甲酸受体α基因探针进行斑点杂交的结果进一步证明HL-60细胞内存在RAR,该受体基因的表达受到RA自身的诱导。
The retinoic acid (RA) receptors in human leukemia cell line HL-60 were studied by radioimmunoassay and molecular sieve HPLC. Found in HL-60 cell nucleus RA-specific binding sites. Scatchard analysis showed that the binding sites were ku 2.03nmol / L and Bmax 1352.6pmol / mg protein. Molecular sieve HPLC analysis shows that the molecular weight of the binding site is about 50ku. These results suggest that RA-specific binding sites present in the nucleus of HL-60 cells have receptor properties. The results of dot blot hybridization of total cellular RNA and retinoic acid receptor alpha gene probe further confirmed the presence of RAR in HL-60 cells. The expression of this receptor gene was induced by RA itself.
